Penicillin G er det foretrukne lægemiddel til Streptococcus pneumoniae, gruppe A, B, C og G streptokokker, nonenterokok gruppe D streptokokker, viridans gruppe streptokokker og ikke-penicillinaseproducerende stafylokokker.

Penicillin G kaliumsalt bruges til at studere murosomer af stafylokokker og den penicillin-inducerede lysis af Streptococcus mutans. Det er stabilt mod hydrolyse vedβ -lactamaser, såsom penicillinaser og cephalosporinaser.
