ELISA

The Human Amyloid Beta 42 (Hu Abeta 42) ELISA kvantificerer Hu Abeta 42 i humant vævskulturmedium, vævshomogenat og cerebrospinalvæske (CSF). Analysen vil udelukkende genkende både naturlig og rekombinant Hu Abeta 42. Metodens princip The Human Abeta 42 solid-phase sandwich ELISA (enzym-linked immunosorbent assay) er designet til at måle mængden af målet bundet mellem et matchet antistofpar. Et målspecifikt antistof er blevet præcoatet i brøndene på den medfølgende mikroplade. Prøver, standarder eller kontroller tilsættes derefter til disse brønde og binder til det immobiliserede (fangende) antistof. Sandwichen dannes ved tilsætning af det andet (detektor) antistof, en substratopløsning tilsættes, der reagerer med enzym-antistof-målkomplekset for at producere et målbart signal. Intensiteten af dette signal er direkte proportional med koncentrationen af mål til stede i den originale prøve. Beta amyloid is a 40-43 amino acid peptide cleaved from amyloid precursor protein by a protease, gamma-secretase. Beta Amyloid (1-40) (Amyloid beta 40 or Abeta 40) together with beta Amyloid (1-42) (Amyloid beta 42 or Abeta 42) are two major C-terminal variants of the amyloid beta protein, constituting the majority of amyloid beta proteins. Both the Amyloid beta 40 and Amyloid beta 42 alloforms are major components of the extracellular plaques found in Alzheimer's disease brain tissue.
