Description: The monoclonal antibody 1-155-2 recognizes the lambda light chain of human immunoglobulin. Approximately 22-kDa, the Ig lambda chain is comprised of one variable and one constant region. Two lambda chains together with the heavy chain comprise the whole Ig structure. Expression of Ig lambda is typically found in less than half of B lymphocytes, and has been associated with neoplastic lymphomas. Applications Reported: This 1-155-2 antibody has been reported for use in flow cytometric analysis. Applications Tested: This 1-155-2 antibody has been pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. This may be used at 5 μL (0.5 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. Super Bright 600 is a tandem dye that can be excited with the violet laser line (405 nm) and emits at 600 nm. We recommend using a 610/20 bandpass filter. Please make sure that your instrument is capable of detecting this fluorochrome. When using two or more Super Bright dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (Product No. SB-4401) to minimize any non-specific polymer interactions. Immunoglobulin classes share the same basic four polypeptide chain structure of two heavy chains (five types) and two light chains (kappa, lambda; both having a molecular weight of 22.5 kDa). Kappa and lambda consist of a variable region and a constant region and can easily be differentiated by the antigenic properties of the constant region. The ratio of kappa to lambda is 70:30. Antibodies to the kappa light chain are reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.
