Description
- Recognizes human P-cadherin
- Cadherins are a family of transmembrane glycoproteins that mediate calcium-dependent cell-cell adhesion and play role in maintenance of normal tissue architecture
- P-cadherin (Placental cadherin, CDH3, cadherin-3) is a classical member of cadherin superfamily
- Expression reported in many cancers (breast, gastric, endometrial, ovian and prostate)
- Can be cleaved into soluble fragment, found in human milk, semen, serum and NAF
Process:
- Antibody specific for P-cadherin pre-coated onto microwells
- P-cadherin protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for P-cadherin is added to detect captured P-cadherin protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
