Description
- Recognizes human N-cadherin
- Cadherins are a family of transmembrane glycoproteins mediate calcium-dependent cell-cell adhesion and plays a role in maintaining normal tissue architecture
- N-cadherin (Neural cadherin, CDH2, Cadherin 2) is a 130 kDa transmembrane protein and classical member of cadherin superfamily
- Expression of N-cadherin reported on various cell types (neurons, endothelial cells, cardiac myocytes)
- Functions as early brain morphogenesis, synaptogenesis, synaptic plasticity
- Soluble N-cadherin (sN-CAD) found in biological fluids (serum, seminal fluid samples, urine)
- Higher amounts of sN-CAD present in cancer patients
Process:
- Antibody specific for N-cadherin pre-coated onto microwells
- N-cadherin protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for human N-cadherin is added to detect captured human N-cadherin protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
