Description
- Recognizes human HER2
- HER2 (ErbB2, Neu) is a member of epidermal growth factor receptor family
- Overexpression of HER2 reported in cancers (breast, ovarian lung, gastric, oral)
- Therapeutic target for treatment of breast cancer and other carcinomas
- HER2 is a 185 kDa transmembrane glycoprotein, extracellular domain of which can be cleaved from cell surface by matrix metalloproteases and released into blood
- Elevated serum levels reported in 23 to 80% of patients with metastatic breast cancer
- Raising serum levels correlated with indicators of poor prognosis and poor response to chemotherapy
Process:
- Antibody specific for HER2 pre-coated onto microwells
- HER2 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for HER2 is added to detect captured HER2 protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
