Description: This B73.1 monoclonal antibody reacts with human CD16, a member of the Fc receptor family also known as Fc gamma RIIIa and Fc gamma RIIIb. This 50-65 kDa receptor is expressed on natural killer cells, neutrophils, monocytes, and macrophages. Crosslinking of CD16 in natural killer cells and monocytes results in activation of the MAPK and PI3K signaling pathways, thus regulating the production of TNF alpha, IL-6, and IL-1 beta. This monoclonal antibody has been shown to block Fc receptor functions. Applications Reported: This B73.1 (B73.1.1) antibody has been reported for use in flow cytometric analysis. Applications Tested: This B73.1 (B73.1.1) antibody has been pre-titrated and tested by flow cytometric analysis of normal peripheral blood cells. This can be used at 5 µL (0.5 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. Excitation: 488-561 nm; Emission: 578 nm; Laser: Blue Laser, Green Laser, Yellow-Green Laser. Filtration: 0.2 μm post-manufacturing filtered.
CD16 encodes a receptor that recognizes the Fc portion of immunoglobulin G and is involved in the clearance of immune complexes from the circulation, as well as other functions such as cellular mediated cytotoxicity and enhancement of virus infections. This gene, FCGR3A, shares a high degree of similarity with another nearby gene, FCGR3B, located on chromosome 1. The receptor encoded by this gene is expressed on natural killer (NK) cells as an integral membrane glycoprotein anchored through a transmembrane peptide, while FCGR3B is expressed on polymorphonuclear neutrophils (PMN) where the receptor is anchored through a phosphatidylinositol (PI) linkage. Mutations in this gene have been associated with immunodeficiency 20 and have been linked to susceptibility to recurrent viral infections, susceptibility to systemic lupus erythematosus, and alloimmune neonatal neutropenia. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. Diseases associated with FCGR3A include Immunodeficiency 20 and Herpes Zoster.Specifications
