Deuterated solvents differ from their non-deuterated counterparts by having one or more of their ordinary hydrogen atoms (protons) replaced with the stable deuterium isotope. This does not alter the chemical properties of the solvent but does confer them with different physical characteristics.

The primary application of deuterated solvents is in NMR, nuclear magnetic resonance spectroscopy (also known as MRS, magnetic resonance spectroscopy). NMR is a technique used to characterise the structure of simple organic molecules through to more complex biological macromolecules. Deuterated solvents are used in NMR for several key reasons:

  • To minimise interference between the sample and solvent signals
  • To help stabilise the magnetic field and avoid drift
  • To more accurately aid system calibration

Most common laboratory solvents are available in their deuterated forms, denoted using the suffix ‘-dx’ (where x = the degree of substitution), for example acetone-d6, acetonitrile-d3, chloroform-d, dimethylformamide-d7, methanol-d4, methyl-sulfoxide-d6, pyridine-d5, tetrachloroethane-d2, tetrahydrofuran-d8, plus others including deuterium oxide, or heavy water.

High isotopic enrichment (typically 99.8% plus, denoted as 99.8atom%D) and high purity (especially low water content) are important factors in NMR, and so most deuterated solvents are manufactured in an inert anhydrous atmosphere and may be supplied in pre-scored ampoules or special septum-sealed bottles to keep them drier for longer after opening. They may contain an internal standard such as TMS (tetramethyl silane), which produces a single, clear signal well away from most other peaks.

Deuterated solvents are sometimes also used in organic chemistry as precursors for the synthesis of other deuterated species.
