Dapper homolog 1, an ortholog of Xenopus Dpr, antagonizes Wnt signaling by promoting Dvl degradation via a lysosome inhibitor-sensitive and proteasome inhibitor-insensitive mechanism. Dapper homolog 1 acts as an interacting protein for Dvl, and the interaction is formed between the DEP (Dishevelled, Egl-10, and pleckstrin) domain of Dvl and the central and C-terminal regions of Dapper homolog 1. The gene for human Dapper homolog 1 is localized on chromosomal region 14q23.1.
